چکیده

INTRODUCTION AND METHODS. Inositol-l,4,5-trisphosphate (IP3) is an important second messenger which mobilizes intracellular calcium stores (1). It has been demonstrated that in various tissues, including the retina, stimulation of several receptor types activates hydrolysis of membrane phospholipids with concomitant formation of IP3 (1-3). Data on effects of dopamine (DA) on IP3 production are scarce and contradictory, and to our knowledge, lacking for avian retina. Some authors reported that activation of a D2 subtype of DA receptors (a member of a D2-DA receptors family) can stimulate IP3 formation, while others did not observe any effect of DA and agonists of DA receptors on phosphoinositide breakdown (2,4,5). This work was aimed at examining effects of DA-ergic compounds on IP3 level in chick retina. Male white leghorn chicks (kept from the day of hatching under 12 h light:12 h dark illumination cycle) were decapitated during the 2nd h of the dark phase. Effects of DA-ergic drugs on IP3 formation were examined in an eye cup preparation. IP3 levels 3 were measured in extracts of retinal supernatants using a specific IP3 [ HI radioreceptor assay kit (Du Pont NEN, Boston, USA). Data are expressed as mean + SEM values (n=7-lO/group). For statistical evaluation of results ANOVA was used followed by Student-Newman-Keuls test. RESULTS AND DISCUSSION. Incubation of chick eye cups with 0.1 pM of bromocriptine (BRC) and quinpirole (QNP), agonists of D2-DA receptors, increased the IP3 level of the retina by 142% and 10896, respectively. (k)-1-Phenyl-2,3,4-tetrahydro-(1H)-3-benzazepine-7,8-diol (SKF 38393; 1 pM), an agonist of Dl-DA receptors, was ineffective. The stimulatory effect of QNP on IP3 formation in chick retina was significantly attenuated by D2-DA receptors antagonists, clozapine (CLOZ) and raclopride (RACL), 0.1 pM each (Fig. 1). To our knowledge, this is the first demonstration showing that stimulation of D2-DA receptors activates formation of IP3 in chick retina. The physiological significance of this phenomenon is yet to be established.

D1 dopamine receptor coupling to PLCβ regulates forward locomotion in mice.

Several studies have reported the coupling of dopamine signaling to phospholipase C β (PLCβ) both in vitro and in vivo. However, the precise physiological relevance of this signaling pathway in mediating dopamine behaviors is still unclear. Here we report that stimulation of dopamine receptor signaling in vivo with systemic administration of apomorphine, amphetamine, and cocaine leads to increa...

Lateral hypothalamus chemical stimulation-induced antinociception was attenuated by injection of dopamine D1 and D2 receptor antagonists in the ventral tegmental area

Introduction: Stimulation or inactivation of the lateral hypothalamus (LH) produces antinociception. Studies showed a role for the ventral tegmental area (VTA) in the antinociception induced by LH chemical stimulation through the orexinergic receptors. In this study, we assessed the role of intra-VTA dopamine D1 and D2 receptors in antinociceptive effects of cholinergic agonist, carbachol, m...

The effect of dextromethorphan on apomorphine-induced pecking behavior in chick

Dextromethorphan is an NMDA receptor antagonist in the glutamatergic system. Currently, there are some reports showing that the glutamatergic NMDA receptor mechanism stimulates dopamine release from several brain regions. This effect may in part modulate the stereotyped behaviors of dopaminergic system. The purpose of the present study was to determine the interaction between the blockade of NM...

Presence of prejunctional D2-dopaminoceptors and α2-adrenoceptors on the cholinergic nerve of the common bile duct of guinea pig

On most adrenergic and cholinergic nerve terminals, prejunctional α-adrenoceptors belonging to the α2-subtype have been identified. Activation of these receptors will decrease the release of norepinephrine. It has been reported that several isolated tissue preparations contain prejunctional dopamine receptors, the stimulation of which inhibits neurotransmission. It has remained uncertain whethe...

Effect of nicotine on immobility time in mice

In the present study, the effects of different doses of nicotine on immobility time in mice were tested. Intraperitioneal administration of low doses of nicotine (0.025 mg/kg) decreased, but higher doses (0.8 and 1 mg/kg) increased immobility time. The anti-immobility response induced by low doses of nicotine was inhibited by high doses of the D2 receptor antagonist sulpiride, the central nicot...
